Dysfunction of actin dynamics after jasplakinolide treatment leads to reduced actin dynamics and reduced chronological life span in DBY746, tor1Δ, sch9Δ, and ras2Δ yeast cells. (A) 2 μM of jasplakinolide was added into the medium. The actin dynamic pattern was examined by fluorescent microscopy. Scale bar, 5 μm. Cells were fixed for 1 h by adding 37% formaldehyde solution directly to growth medium to a final concentration of 3.7%. After washing with 1× PBS solution for three times, cells were stained with phalloidin-TRITC in the dark at room temperature for 1 h and images were taken by fluorescent microscope.
