From: APP family member dimeric complexes are formed predominantly in synaptic compartments
The dimer amount is independent of different APP splice variants. A APP695 HA, APP751 HA and APP770 HA were heterologously expressed in N2a cells. Non-transfected N2a cells served as a negative control. Equal amounts of cell lysates were separated on SDS-Gels and Western blot detection followed with an α-HA antibody. B APP695 HA, APP751 HA and APP770 HA or APP F1 were heterologously expressed in N2a cells. To show APP dimers, APP F1 expressing cells were treated with 100 nM of the rapamycin analogue AP20187 overnight to induce dimerization of APP. Incubation with the solvent of AP20187, ethanol, served as a negative control. The samples were analyzed via Blue Native PAGE and Western blot analysis followed with an α-HA antibody
