SARS-CoV-2 NSP13 prevents nuclear translocation of STAT1 and STAT2. A NSP13 blocks IFN-β- and IFN-γ-induced nuclear translocation of STAT1. A549 cells were transfected with an expression plasmid for V5-tagged NSP13. At 24 h post transfection, transfected cells were stimulated by 1000 U/mL IFN-β or 100 ng/mL IFN-γ for 30 min, and then stained for total STAT1 (red) and V5 (green). 4′, 6-diamidino-2-phenylindole (DAPI, blue) was used to visualize nuclear morphology. Transfected cells were highlighted by arrows. STAT1 and DAPI signals were merged in panels 2, 5, 8 and 11, while STAT1, NSP13 and DAPI signals were merged in panels 6 and 12. B NSP13 counteracts IFN-β-induced nuclear translocation of STAT2. Endogenous STAT2 was in red.
