MDLS promoted the expression of pluripotency markers in the expansion of mESCs. OG2-mES cells were plated onto gelatin-coated six-well plates at a density of 1 × 104 cells/well and cultured with 5 μg/mL MDLS, BMP4 or DMSO without feeder cells. They were split every 3 days and seeded at the same density. A Left: the expression of Oct4, Sox2 and Nanog in the OG2-mES cells treated by MDLS at passage 3 was detected using Western blot analysis. Right: quantitative analysis of Western blotting data by image J soft-ware. Levels of these proteins were normalized to the corresponding GAPDH band expressed as relative fold changes in comparison to control samples.
