MDLS enhanced the self-renewal of mESCs. A The cytotoxic effect of MDLS on OG2-mES cells. OG2-mES cells were treated with different concentrations (2.5, 5, 10, 20, and 40 μg/mL) of MDLS for 44 h and MTT assay was conducted. The results were shown as mean ± SD of three separate experiments. B The role of MDLS in the self-renewal of OG2-mES cells in the absence of feeder cells and LIF. Undifferentiated OG2-mES cells were plated onto gelatin-coated black 96-well plates at a density of 1 × 104 cells/well without feeder cells. After 24 h, the medium was replaced by fresh medium with or without LIF but containing 5 μg/mL MDLS or DMSO. After another 6 days of incubation, self-renewal of mESCs was examined by analyzing GFP fluorescence intensity.
