Knock-down of PD-L1 triggered apoptotic and pyroptotic cell death in cisplatin treated CR-GC cells in vitro and in vivo. The expression levels of a, b PD-L1 and NLRP3, and c, d pyroptosis associated biomarkers were evaluated by Western Blot analysis. e The Annexin V-FITC/PI double staining assay was performed to determine cell apoptosis ratio. Western Blot analysis was conducted to detect the protein levels of f, g Cyclin D1 and CDK2, and h, i cleaved Caspase-3 and Bax in mice tumor tissues. (Note: “Con” represented “Control”, “OE” indicated “PD-L1 overexpression”, “KD” represented “PD-L1 silence”, “Cis” indicated “Cisplatin”, “C + K” represented “Cisplatin plus PD-L1 ablation”, “C + K + K” represented “Cisplatin plus PD-L1 ablation plus silencing NLRP3”). Each experiment repeated at least three times. *P < 0.05
