The role of VCAN in HG-HUVEC-Exos-induced VSMCs senescence/calcification. HUVECs were transfected with negative control siRNA (con-siRNA) or VCAN siRNA (VCAN-siRNA) for 6 h and then exposed to HG condition for 48 h. After that, exosomes were isolated and co-cultured with VSMC for 48 h. The protein expression of ALP, p21, HADHA and Cox-4 were examined by Western blot. The ALP, p21, HADHA and Cox-4 levels were quantified and normalized to GAPDH.
